Principle of the method
The PCL-method is based on the photo-induced peroxide-radical-induced autoxidation of luminol. By optical excitation of the photosensitizer luminol measuring radicals, especially peroxide-anion-radicals are generated which partly are eliminated by the reaction with antioxidants in the sample. The chemo-luminescence of a detector substance produced by remaining radicals is measured in a measuring cuvette and thus the quantity of antioxidants in the sample quantified. The integral antioxidative capacity thus determined is related to equivalent concentration units of the used standards ascorbinic acid and trolox as tocopherol analogon.
Evaluation
The results reveal that the analyzed sample FAME "Completec“ has antioxidative radical-scavenging properties and is able to capture and to neutralize peroxide-anion-radicals generated in this test system.
The radical-scavenging activity of the product FAME "Completec“ amounts to 70.0 µmol ascorbic acid equivalent per sample in the aqueous test system (ACW). In the lipid system (ACL) the effective activity is comparable with 289.4 µmol trolox units. The sum of the antioxidative radical-scavenging effective activity can be characterized by 359.4 µmol/g total AO-equivalents.
Further, the results of the antioxidative activity of 13 reference antioxidants determined by the PCL-method are given in mmol/mmol and µmol/g.
The highly efficient antioxidant and radical scavenger vitamin C (ascorbic acid) had the highest efficient activity in scavenging the aggressive peroxide radicals, in the PCL test system. The radical-scavenging activity of vitamin C amounts to 792.6 mmol ascorbic acid equivalents per mmol in the aqueous test system (ACW), in the lipid system (ACL) the activity is comparable with 1,373 µmol trolox units.
The natural product pycnogenol also had very good radical-scavenging properties in the aqueous as well as in the lipophilic test systems (activities of 627 µmol/g and 5,250 µmol/g).
The tested carotenoids astaxanthin and beta-carotene had comparably low radical-scavenging activities against peroxide-anions. However, it is quite possible that in other test systems and against other reactive radicals or oxygen species, e.g. singlet oxygen, these substances have higher antioxidative activities.
